Goal and objectives of the project | Culturing and stimulating proliferation of umbilical cord hematopoietic stem cells using peptide SL-13R. |
Main results | (1) The mononuclear blood cell population was obtained by ficoll-Paque gradient centrifugation, and umbilical cord blood stem cells were isolated from the mononuclear cell population after the mononuclear blood population was enriched with UC-HSC cells for 3 weeks. The obtained UC-HSC population had 91.94% CD34 positive cells. (2) The UC-HSC cells were tested for proliferation stimulation by SL-13R peptide at a concentration of 10µg/ml over time, the proliferation rate was 1.21 to 1.44 times greater than that of the unstimulated control sample. (3) The UC-HSC cells stimulated to proliferate by SL-13R peptide were tested for CD34 positive percentage, the results were UC-HSC1 93.73%; UC-HSC2 95.94%; UC-HSC3 93.76% higher than the unstimulated control sample UC-HSC1 91.94%. SL-13R peptide stimulated proliferation and maintained the stemness of umbilical cord blood stem cells well. (4) Testing the concentration of SL-13R and KS-13 peptides on proliferation stimulation of mesenchymal stem cells (AD-MSC) showed that the proliferation rate was proportional to the concentration of the proliferative peptide, the concentration above 10μg/ml, the proliferation rate did not have a big difference. These results demonstrate that both SL-13R and KS-13 peptides enhanced AD-MSC proliferation compared to the control group on day 3. |
Novelty and actuality and scientific meaningfulness of the results | Successfully isolated and cultured hematopoietic stem cells from umbilical cord blood and mesenchymal stem cells from adipose tissue and tested the stimulation of proliferation of both types of stem cells by peptide SL-13R. |
