Goal and objectives of the project

- Isolation and structrural elucidation of chemical constituents of Platycodon grandiflorus.
 - Evaluation of hypolipidemic effects of the isolated compounds

Main results

- Seventeen compounds (PG1-PG17) were isolated from the methanolic extract of Platycodon grandiflorus roots. Their structures were elucidated by means of spectroscopic methods, including 1D, 2D NMR and MS, and comparison their spectra data with those reported in literature. Among them, two new compounds were obtained.
- Fifteen compounds (PG1-PG15) were investigated on hypolipidemic effects. Compounds PG10, PG11, PG8, PG1 and PG2 showed the capacity of reducing intracellular lipid levels on the HepG2 cell line.
-  Compound α-ethenyl-4-hydroxy-3,5-dimethoxybenzenemethanol (PG11) was further studied on hypolipidemic effect. This compound showed cytoprotective effect against H2O2-induced cell death through suppressing the gene expression of Keap-1 and inducing the expression of Nrf-2, HO-1 and NQO-1 genes, which involved in antioxidant defense. In addition, increasing the expression of CYP7A1 and decreasing of the expression of HMG-CoA reductase, FAS and SCD-1 are the main reason for hypolipidemic effect of PG11.

Novelty and actuality and scientific meaningfulness of the results

- Two new compounds were isolated from Platycodon grandiflorus roots.
- This is the first report on the hypolipidemic effect of compound α-ethenyl-4-hydroxy-3,5-dimethoxybenzenemethanol (PG11).
