Goal and objectives of the project

Evaluation of the biosynthetic potential of neuroprotective and cardioprotective substances of filamentous fungi from Khanh Hoa sea for application in medicine.

Main results

- Screening of 15/35 strains (42%) capable of reducing DPPH and ABTS free radicals with SC% value > 50%. 
- Screening of 23/35 (65%) fungal strains capable of inhibiting less than 50% the survival of H9c2 cardiomyocytes cells and 7/35 (20%) of Neuro-2a cells. 
- Screening of 21/35 (60%) fungal strains capable of inhibiting more than 50% the growth of Hela cervical cancer cells and 16/35 (46%) for MCF-7 breast cancer cells.
- Classification of 04 selected fungal strains with high bioactivity was Penicillium sp. 1901NT-3.7.1, Penicillium sp. 1901NT-3.9.1, Aspergillus terreus 1901NT-3.19.1 and Aspergillus carneus KMM 4638.
- Isolation of 04 compounds including asperflavinoid C (1), strobilactone A (2), ustusolate E (3), (6-strobilactone A) ester of (E, E)- 6-carbonyl-7-hydroxy-2,4-octadienoic acid (4) from a coculture extract of the marine fungi A. carneus KMM 4638 and B. felina KMM 4639.
- Strobilactone A (2) exhibited protective activity towards H9c2 cardiomyocyte cells when treated with rotenone. Asperflavinoid C (1), ustusolate E (3), and (6-strobilactone A) ester of (E,E)-6-carbonyl-7-hydroxy-2,4-octadienoic acid (4) showed cytotoxicity against human cancer cell lines including breast cancer MCF-7, prostate cancer PC-3, and leukemia Raji.

Novelty and actuality and scientific meaningfulness of the results

The study isolated 04 compounds from the co-culture extract of the fungi Aspergillus carneus KMM 4638 and Beauveria felina KMM 4639, of which strobilactone A (2) was also evaluated for the first time with protective activity towards H9c2 cardiomyocyte cells when treated with rotenone, asperflavinoid C (1) and ustusolate E (3) were evaluated for the first time with cytotoxic activity against MCF-7 breast cancer cells.

Recommendations

Conclusion:
Scientists from the Pacific Institute of Bioorganic Chemistry (PIBOC-FEB-RAS) are the world's leading scientists in the field of bioactive compounds from marine organisms for functional foods and new medicines for treating diseases. Therefore, the cooperation with Russian scientists is extremely advantageous to deploy this new research direction in our country. The research results of the Mission, in addition to producing new scientific and technological data, also contribute to improving the professional qualifications of scientists in researching new issues of marine science and technology.
Suggestion:
It is necessary to continue expanding the direction of researching bioactive substances from microorganisms in Vietnam's waters to rationally exploit, use and conserve marine natural resources, along with affirm the biodiversity potential of our sea area.
