Goal and objectives of the project

Study on chemical composition and antibiotic, anti-cancer activities of Zanthoxylum rhetsa, orientation to make preparations for medical and pharmaceutical applications.

Main results
About the science
- The project has implemented all the research contents registered in the approved contract, including:
+ Collected the samples of Zanthoxylum rhetsa (leaves, fruits, stem barks), sample collection records were made.
+ Processed samples and created fractional extracts from bark and leaves, and collected essential oils from fruit.
+ The chemical composition of essential oils (EO) has been determined by GC-MS method. The results obtained are as follows:
¬ For fresh fruits EO: white, aroma, lighter than water, 7.4% compared to the raw material. Physical parameters: density 0.8888, refractive index 1.4770, polar rotation 43.88. Chemical composition: the main components are monoterpenes (98.75%), sesquiterpenes only account for a small amount (0.24%). Among the monoterpenes, sabinene was highest (41.13%), followed by terpinolene (27.05%), limonene (7.30%), α-terpineol (6.08%), terpinen-4-ol (5.35%). Other components trans-β-ocimene, γ-terpinene are also present but in lower concentrations (1-2%).
¬ For dried fruits EO: white, aroma, lighter than water, 9.4% compared to the raw material. Physical parameters: density 0.8390, refractive index 1.4780, polar rotation 75.09. Chemical composition: the main components are monoterpenes (99.02%), sesquiterpenes only account for a small amount (0.38%). Among the monoterpenes, sabinene was highest (33.71%), followed by terpinolene (30.37%), limonene (8.29%), terpinen-4-ol (7.72%). Other components α-terpineol, myrcene, α-terpinene, trans-β-ocimene are also present but in lower concentrations (2-3%).
+ The chemical composition of the steam bark of Z. rhetsa has been studied. As a result, 15 compounds were isolated and determined: nitidine (ZR-V01), 6-acetonyldihydronitidine (ZR-V08), 6-acetonyldihydrochelerythrine (ZR-V07), zanthorhetsavietnamese (ZR-V10), isocorydine (ZR-V03),  (O)-methyltembamide (ZR-V14), (N-(4-methoxyphenethyl)benzamide (ZR-V15), piperonylic acid (ZR-V09), asarinin (ZR-V04), horsfieldin (ZR-V13), 5-(4-(3,4,5-trimethoxyphenyl)hexahydrofuro[3,4-c]furan-1-yl)benzo[d][1,3]dioxole (ZR-V17), 5-(4-(3,5-dimethoxyphenyl)hexahydrofuro[3,4-c]furan-1-yl)benzo[d][1,3]dioxole (ZR-V19), hesperidine (ZR-01), syringin (ZR-V21), β-sitosterol (ZR-V06), in which, Zanthorhetsavietnamese (ZR-V10) is a new compound.
+ The chemical composition of the leaves of Z. rhetsa has been studied. As a result, 08 compounds were isolated: 3-O-beta-D-glucopyranosyl-beta-sitosterol (ZR-L01), 1,4-bis(4-methoxyphenyl)hexahydrofuro[3,4-c]furan (ZR-L03), asarinin (ZR-L05), 5-(4-(3,5-dimethoxyphenyl)hexahydrofuro[3,4-c]furan-1-yl)benzo[d][1,3]dioxole (ZR-L06), (1R,4S)-1,4-bis(3,5-dimethoxyphenyl)hexahydrofuro[3,4-c]furan (ZR-L10), (Z)-methyl 3-(3,4-dihydroxyphenyl)acrylate (ZR-L08), 4-hydroxybenzoic acid (ZR-L11), acid gallic (ZR-L04).
+ Evaluated the antifungal, antibacterial, cytotoxic activities of the obtained compounds and EO: 
¬ For antifungal and antibacterial activities: 11/17 samples showed activity ZR-V01, ZR-V08, ZR-V07, ZR-V01, ZR-V03, ZR-V15, ZR-V09, ZR-L08, ZR-L11, fresh fruit EO, dried fruit EO. In which, the fresh fruit EO is active against E. coli, F. oxysporum, S. serevisiae (MIC 100 µg/mL).
¬ For cytotoxic activity: 12/17 samples showed activity ZR-V01, ZR-V08, ZR-V07, ZR-V10, ZR-V03, ZR-V15, ZR-V09, ZR-01, ZR-L08, ZR-L11, fresh fruit EO, dried fruit EO. In which, nitidine (ZR-V01) was very active, inhibiting all three cell lines (IC50 values ​​lower than Elippticine); both samples of fresh and dried fruits EO showed good activity against all three tested cancer cell lines.
- In addition, the thesis has also implemented some research contents that are not registered in the contract. These are the following: 
+ Isolated and determined the chemical structure of some secondary compounds in the fruits of Z. rhetsa (05 compounds). The main components of the fruits are fatty compounds.
+ Studied on the chemical composition and biological activities of essential oils obtained from some parts of Z. rhetsa: leaves, twigs, fruit branches, bark trunks.
+ Studied on the chemical composition and antifungal, antibacterial, cytotoxic activities of Z. rhetsa EO collected in two different localities: Quan Son, Thanh Hoa and Thuan Chau, Son La.
Novelty and actuality and scientific meaningfulness of the results

- Studied on the chemical composition of leaves, fruits, bark of Z. rhetsa (total 22 isolated compounds, including 01 new compound). 
- Studied on the chemical composition and antifungal, antibacterial, and cytotoxic activities of essential oil obtained from the fruit, leaves, twigs, twigs and barks of Z. rhetsa.

Recommendations

We would like to propose Vietnam Academy of Science and Technology and Institute of Natural Products Chemistry to help us to be continuously investigated to carry out develop these compounds for pharmaceutical industries to produce a medication product in order to strengthen public health.
Nitidine and fruits essential oil accounts for high content in Zanthoxylum rhetsa, that have good antifungal, antibacterial and cytotoxic activities. Therefore, there should be more studies on some other activities (anti-inflammatory, analgesic, antioxidant, immune-enhancing...), oriented to develop into natural products applied in medicine and pharmacy.
