Goal and objectives of the project

Identification and evaluation of distribution, biological activities (antibacteria and anticancer) of endophytic actinomycetes from mangrove plants (Bruguiera gymnorrhiza, Avicennia marina, Agiceras corniculatum) collected in Quang Ninh province.

Main results

Theoretical results:
A total of 62 endophytic actinomycetes was isolated from Avicennia marina, Agiceras corniculatum, and Bruguiera gymnorrhiza collected in Quang Ninh province.
Out of 62 endophytes, strain SX6 from A. corniculatum and MX9 from B. gymnorrhiza showed significant antibacterial activity against at least 4 tested bacteria with inhibition zones ranging from 7,96-32,5 mm. Besdies, ethyl acatate extract of both strains also displayed cytotoxic activity against 3 cancer cell lines including A549, MCF7, and Hep-3B.
Strains SX6 and MX9 were identified as S. parvulus SX6 and S. parvulus MX9, respectively. S. parvulus MX9 was deposited at VAST-Culture Collection of Microorganisms under accession number VCCM 22513.
S. parvulus SX6 and S. parvulus VCCM 22513 were sequenced by Illumina platform in order to study genetic basis involved in antibacterial and anticancer properties. The genome of S. parvulus SX6 contained 04 novel gene clusters atttributed to antibacterial and anticancer activities. In addition, biosynthetic pathways of plant-derived compounds such as daizdein and genistein were proposed for the first time. On the other hand, adaptation of S. parvulus VCCM 22513 to endophytic lifestyle was demonstrated using genome mining.
Applied results: S. parvulus SX6 could be an effective platform for daizdein and genistein production which could be applied in medicinal and pharmacetutical industries. These findings promote employing whole-genome sequencing in finding new metabolites with biological activities. 

Novelty and actuality and scientific meaningfulness of the results

The project exploited for the first time biosynthetic pathways of daizdein and genistein from endophytic S. parvulus SX6.The underlying mechanisms involved endophytic lifestyle of S. parvulus VCCM 22513 from from B. gymnorrhiza were proved for the first time. These could serve as reference genomes for domestic and international studies.
