Goal and objectives of the project

- Evaluate the content of chlorogenic acid in green coffee beans collected in some provinces of the Central Highlands of Vietnam to select raw materials for extraction.
- Developed a technological process for extracting green coffee powder containing total chlorogenic acid content of CGAs 10-15% and 40-50% with a scale of 30 kg of dry materials/batch, which meets export standards.
Developed a process to produce the standard 3-CQA 98% at laboratory scale.

Main results

Theoretical results:
+ Six pure compounds have been isolated from green coffee beans: 5-O-caffeoyquinic acid (1), 3-O-caffeoyquinic acid (2), 4-O-caffeoyquinic acid (3), 3,5-O- dicaffeoylquinic acid (4), 3,4-O-dicaffeoylquinic acid (5) and 4,5-O-dicaffeoylquinic acid (6).
+ Developed qualitative and quantitative method of chlorogenic acids from green coffee beans using HPLC and LC-MS Qtop.
+ Evaluated the total chlorogenic acid content in 24 raw green coffee samples collected in some growing areas in Vietnam, reaching about 4.9-6.5%. Robusta coffee has a higher content of CGAs than other types of coffee. The content of CGAs in coffee silk (a by-product of roasting coffee processing) is about 2.0-2.1%, in dry coffee leaves about 1.3-1.5%.
+ Developed a technological process to extract green coffee powder with 10 - 15%  and 40-50% of CGAs in a scale of 30 kg of raw materials/batch. The efficiency was about 29.33% and 8.4% compared to the dry material sample weight. Developed a purification process for chlorogenic acid (3-CQA) over 98% from green coffee powder 40-50% of CGAs with a scale of 50 grams/batch.
+ Developed basic standards for green coffee powder with chlorogenic acid content of CGAs 10-15%, CGAs 40 -50% and CGA 98%.
+ Conducted trial production of the products in a pilot scale of 30 kg of raw materials/batch. Total of 26.44 kg of green coffee powder with about 15% of CGAs and 7.54 kg green coffee powder with about 50 % of CGAs was produced.
+ Researched to create powder to support weight loss and evaluated the preclinical pharmacological effects of the product. The results show that the product CFX 40 is safe (no acute toxicity in experimental animals, rats, orally at a maximum dose of 10,000 mg/kg body weight). Powder for weight loss - CFX 40 at a test dose of 250mg/kgP has anti-obesity and anti-hyperlipidemia effects by reducing body weight and serum lipid levels.

Novelty and actuality and scientific meaningfulness of the results

The obtained results are new findings in Vietnam. The production processes of green coffee powder with 10-15% of CGAs and 40-50% of CGAs give good performance, can be applied to produce green coffee preparations for medicinal purposes from domestic green coffee beans.

Recommendations

Need to continue testing time 2-3 months on a larger number of people (50-100 people) to evaluate the product's effectiveness and compare with similar products on the market.
