Goal and objectives of the project

- Simulation to screen the compounds that are able to inhibit ecto-5'-nucleotidase (CD73) enzyme.
- Synthesis of potential pyrophosphate derivatives based on the screening results.
-  Evaluation of biological activity of synthezied compounds with CD73 enzyme.

Main results

Theoretical results:
+ Designed and screened the novel pyrophosphate derivatives in silico to select potential targets of CD73 for synthesis.
+ 4 novel compounds were synthesized: [1H-1,2,3-triazole-4-(1-methoxy-3-methyl-9H-carbazole)]phosphinyl-methylphosphonate (trisodium salt) (12), [1H-1,2,3-triazole-4-(4-methoxycoumarin)phosphinyl]-methylphosphonate (trisodium salt) (13), [1H-1,2,3-triazole-4-(10-methylcaridin-9(10H)-one)phosphinyl]-methylphosphonate (trisodium salt) (14) and [1H-1,2,3-triazole-4-(1-methoxy-3,9-dimethyl-9H-carbazole)phosphinyl]-methylphosphonate (trisodium salt) (15).
+ The obtained compounds 12-15 were evaluated with CD73 enzyme. The results showed that compound 13 was moderately inhibitory to CD73.
+ Used silico model to explain the biological activity results.

Novelty and actuality and scientific meaningfulness of the results

This project has synthesized the novel pyrophosphate compounds and evaluated their CD73 enzyme inhibitory activites.

The project has achieved some initial results on the synthesis of pyrophosphate compounds and their inhibitory ability to the CD73 enzyme. However, futher studies of pyrophosphate derivatives are needed to find a stronger inhibitory of CD73.
