Goal and objectives of the project Investigation of anti-proliferative and apoptosis induction potential of Moringa Oleifera Lam.leaf extract against other melanoma cell lines.
Main results

- The crude extract and fractions derived from methanol extract, such as n-hexane fraction, chloroform fraction, ethyl acetate fraction and water-soluble fraction of Moringa oleifera leaves were successfully collected. Total phenolic and flavonoid contents of extracts or fractions were determined. The total flavonoid content is positive correlation with total phenolic content in extracts or fractions. As a result, ethyl acetate fractioncontained noticeable amount of phenolic acids and flavonoids.
- Methanol extract andethyl acetate fraction inhibited the proliferation of both human melanoma A375cells and A2058 cells, but had little effect on WS1 normal human skin fibroblasts and primary normal human dermal fibroblasts (NHDF).
- The phenolic-rich ethyl acetate fraction markedly inhibited the growth of A375 cells in a dose- and time-dependent manner. The anti-proliferation was supposed to be mediated via apoptosis, which was demonstrated by the significant increase of condensed chromatin, DNA fragmentation and PS externalization.The phenolic-rich ethyl acetate fraction exerted significant anti-cancer effects on melanoma cells in vitro which involved in both Caspase-dependent and Caspase-independent apoptosis pathways.
- These results provided a fundament for the using of extracts of Moringa oleifera leaves to treat skin cancer effectively.

Novelty and actuality and scientific meaningfulness of the results

The extract from the leaves of Moringa Oleifera Lam. leaves possessed high anti-cancer property against melanoma cells. The mechanism of cell death involved in both Caspase-dependent pathway through activation of Caspase-9 and Caspase-3/7 and through Caspase-independent pathway via AIF translocation.

Products of the project

Technological products: methanol extract, aqueous extract, n-hexane fraction, chloroform fraction, ethyl acetate fraction derived from Moringa oleifera leaves was prepared and characterized. The data for the cytotoxic effect of Moringa oleifera leaf extract against melanoma cells. They were kept at Graduate University of Science and Technology, Vietnam Academy of Science and Technology.
