Identification toxins in some reef fish and other species which potential causing food poisoning in Vietnam for seafood safety.

Main results

- Theoretical results:
+ Ciguatoxin-1B and 2 derivaties including 54-deoxyCTX-1B và 52-epi-54-deoxyCTX-1B was identified by LC/MS-MS analysis in the specimens of red snapper Lujanus bohar causing 02 posoning icidents in Vietnam 2014 and 2016. Together with analysis of CFP identical symptoms, the result affirmed that CTX-1B was causative toxin in CFP case in Vietnam. This is the first report in Southeast Asian countries. On the other result, the study detected the presence of CTX-1B randomly in a certain number of specimens of some morey eels specíe collected from Central of Vietnamese waters with equivalent levels to that detected in snapper poisonous specimens. It is indicated that CFP in Vietnam is potential by consumption of morey eels. Nothiwordly, with a certain level of CTX-1B detected in L. bohar specimens collected from Ninh Hai-Ninh Thuan water, this snapper is considered as a very dangerous species causing CFP in Vietnam.
+ As the first report in Vietnam, tetrodotoxin (TTX) was confirmed in 03 marine snail species included Nassarius glans, N. conoidalis and N. pullus collected from Khanh Hoa water. The result indicated that TTX is causative toxin in reproted poisoning cases by consumption of marine snails in Vietnam. With 100% specimens contained TTX in the levels beyond consumtable limit (10 MU/g) recomended in Japan in term of TTX, the result concluded that N. glans, N. conoidalis và N. pullus are toxic species, not safe to eat. Especially, N. glans is very high toxicity, and may cause human poisoning with 12-13 g (equivalent to 5-6 specimens). By the results, these marine snail species are additional listed into marine toxic animals in Vietnam.
- Applied results:
Practical protocol for CTXs analysis using Shimadzu 8040 LCMS is developed successfully in the VAST keylab on seafood and environmental safety (central of Vietnam) in Institute of Oceanography. By using published method by Japanese scientists, the study had modified some neccesary factors such as collision energy and column temperature. In the practical protocol, CTSs can be isolated in Poroshell 120 EC-C18 (100 × 2,1 mm i.d.; 2,7 mm particle size) solid phase, mobil phase of disstilute water and methanol containing 5 mM ammonium formate and 0,1% linear gradient program with B concentration from 80% to 95% in 10 mins, keep in 10 following mins, flow rate of 0,25 mL/min. Selected scan mode SIM with [M+Na]+ as mother ion and prodcution ions; IS 5500 V, TEM 500 oC, collision energy 40 eV, DF 400 V is suitable to detect and quantify CTXs for delettion limit of CTX-1B (0,01 ppb) in muscle of reef fish with LOD S/N > 3 và S/N > 10 is 0,25 pg and 1,0 pg in column.

Novelty and actuality and scientific meaningfulness of the results

- To contribute in science 2 scientific publications about toxins in snappẻ and marine snails in Vietnam.
- Developed practical protocol has been applied in VAST keylab for curent researches.
- To affirm ability of Vietnam in WESTPAC region about marine toxin study, by support other member states in capacity building and identifiation of causative toxins in poisoning cases when occurred in the region.
