Main results

- Theoretical results:
From the leaves of Markhamia stipulata var. canaense V.S. Dang, three new cycloartane-type triterpenoids, named Markhacanasin A (MS04), Markhacanasin B (MS05), and Markhacanasin C (MS09), together with eight known compounds, oleanolic acid (MS01), ursolic acid (MS02), 6β,19α-dihydroxyursolic acid (MS03), luteolin (MS06), apienin 7-O-β-D-glucopyranoside (MS07), and luteolin 7-O-β-D-glucopyranoside (MS08), naringenin (MS10), tectoquinone (MS11), mollic acid (MS12) and 2,3-dihydroxypropyl palmitate (MS13) were isolated by variously chromatographic methods. All isolated compounds were reported the first time from this species. Their structures were elucidated by IR, UV, HR-ESI-MS and NMR experiments. The cytotoxicity of all triterpenoids (MS01-MS05) against three human cancer cell lines (MCF-7, HepG2, and HeLa)  and the α-glucosidase inhibition of flavonoids (MS07, MS08) were evaluated. At the concentration of 100 μg/mL, MS04 exhibited moderate cytotoxic activity against three test cells (93.93, 91.25, and 92.74 %, respectively), and MS02 showed meaningful cytotoxic effect against HeLa (86.36 %), whereas, MS07 displayed stronger α-glucosidase inhibitory activity (IC50 = 103.76 µg/mL) than the standard drug acarbose (IC50 = 134.13 µg/mL).

 

Novelty and actuality and scientific meaningfulness of the results

For the first time, the phytochemical and biological investigations of Markhamia stipulata var. canaense V.S. Dang were conducted. The systematically investigation of Markhamia stipulata var. canaense V.S. Dang, a new species uncovered in Viet Nam was detailed for the first time from sample collection, sample identification, isolation, structure elucidation and evaluation of cytotoxic and α-glucosidase inhibitory activities of isolated compounds. The data of isolated compounds from this species will update the published database word wide, with the research results of the genus Markhamia will contribute to clarify the pharmaceutical effects of Markhamia species in Vietnamese traditional medicine in the future.
