Goal and objectives of the project

Development of V. amigdalina in Thua Thien Hue and orientation of using V. amigdalina in medicine for economic development and community health care.
Specific objectives:
+ Determine the chemical composition of the medicinal plant from V. amigdalina leaves and build fingerprint chromatography data for themedicinal plant.
+ Evaluating the biological activity of extracts of V. amigdalina leaf (activity of reducing blood glucose, increasing liver function and detoxification, antibacterial activity ...) and screening the main active ingredients.
+ Proposing a trial planting model and orientations for development of the medicinal plant V. amigdalina in Thua Thien Hue
8. Main results

Main results

Theoretical results:

  • Contents 1: Experimental planting of V. Amigdalina in Thua Thien Hue:
    The trial planting of the plant V. Amigdalina in Phong Dien, Phu Vang and Phu Loc showed that the plant were grown and developed at all three trial planting areas. However, the plant at Phong Dien are poorly grown and have low leaf biomass productivity. Therefore, it is not recommended to develop the plant in Phong Dien. At Phu Vang and Phu Loc, the plant is similar well-grown, with no difference between these two regions. Hence, it is possible to grow the plant in Phu Vang and Phu Loc.
  • Content 2: Study the chemical composition of medicinal samples
    From leaves of V. amgydalina, we isolated and identified six compounds belonging to the class of C29-steroid, and established baseline standards for the main active ingredients. This is the characteristic layer of Vernonia amydalina, including:
  1. A new compound, (22R,23S,24R,28S)-3β-glucosyl-28-methoxy-7,8,9,11-tetradehydro-21,24-dihydroxy-21,23:22,28-diepoxy-5α-stigmastane (VA7), is named as vernomyosit E
  2. Two compounds were first isolated from nature (22R,23S,24R,28S)-28-methoxy-7,8,9,11-tetradehydro-3β-16α, 21,24-trihydroxy-21,23:22,28-diepoxy-5α-stigmastane (VA4), namely vernomyosit C and (23S,24R,28S)-3β,22α-dihydroxy-7,8,9,11-tetradehydro-24.28- epoxy-5α-stigmastane-21.23 carbolactone (VA11), namely vernomyosit C1.
  3. Three other known compounds are vernoniacum B (VA5), vernonioside B1 (VA9), vernonioside B2 (VA10).
    Thus, number of isolated compounds are double compared to registered those in approved proposal.
  • Content 3: Building the chromatographic fingerprint data for the medicinal materials
    - Determined the analysing conditions, created chromatographic fingerprint data, and built the calibration curves for quantifying 5 main compounds isolated from the plant in Thua Thien Hue.
    - Quantified and compared the amount of active ingredients in the medicinal samples of samples grown in different locations. The highest concentration of reference materials was found from samples grown in Phu Vang: VA5 = 0.56 µg/mL and VA10 = 0.27 µg/mL
  • Content 4: Evaluate the biological activity of leaf extracts
    a, Evaluate in vitro activity of leaf extract
    - Cytotoxic activity of the leaf extract was assessed on two cell lines: Hep3B and MCF-7 cells. Evaluation results show that the extract exhibit cytotoxic activity against the cell line Hep3B and MCF-7 with IC50 values of 114.82 ± 2.13 and 123.03 ± 1.27 µg/mL, respectively.
    - The methanol extract of the plant leaves exhibit anti-inflammatory activity through inhibition of NO production induced mouse macrophage cells RAW264.7 at a concentration of 100 µg/mL with an inhibition of 61,56 ± 0 ,66%.
    - The methanol extract of the plant leaves showed α-amylase inhibitory and α-glucosidase inhibitory activity with IC50 values of 0.64 ± 0.12 and and 1.78 ± 0.31 mg/mL, respectively.
    b, Evaluation of in vitro activity of isolated substances
    Three puried compounds VA5, VA7, VA10 were tested for of α-amylase and α-glucosidase inhibitory activity. The compound VA7 showed α-glucosidase inhibitory activity more strongly than control acarbose and also exhibited α-amylase inhibitory activity. Compounds VA5 and VA10 showed moderate α-amylase inhibitory activity with IC50 values of 83.17 µg/mL and 87.09 µg/mL respectively.
  • Content 5: Building a trial planting model of V. Amigdalina in Thua Thien Hue
    Built a trial model for growing V. Amigdalina in Phu Vang, Thua Thien Hue.
    - The plant was grown well and gave high leaf biomass yield of 43.4 tons/ha for cut trees after 3 months of planting and 30.4 tons/ha for non-cutting trees, increasing 142.76% compared with non-cutting trees.
    - The extract of the plant leaves exhibited good α-glucosidase inhibitory activity at a concentration of 2 mg/ml and good α-amylase inhibitory activity at a concentration of 1 mg/mL.
    - The extract of the plant leaves grown in Phu Vang reduced blood glucose levels in experimental mouse at a dose of 400 mg/kg/day.

Applied results:
Novelty and actuality and scientific meaningfulness of the results: The project studied growth ability of V. Amigdalina in Thua Thien Hue for the first time and the plant was grown well. Also, the project isolated compounds from the plant leaves including three new compounds. Additionally, biological activities of extract and isolated compounds were were tested.
