| Goal and objectives of the project |
- To search bioactive compounds from some soft corals in Northeast Vietnam sea
- To select the soft coral species with anticancer components or antimicrobial components for preparing the trial product
|
| Main results |
Theoretical results: - 41 samples were collected from research area, 17 soft corals species were identified including three group as Sarcophyton, Sinularia and other species;
- The Sarcophyton pauciplicatum exhibited anticancer activity with cell lines as MCF7 (IC50 = 9.57 µg/ml); LNCaP (IC50 = 5.08 µg/ml); HL-60 (IC50 = 8.00 µg/ml); Hep G2 (IC50 = 5.48 µg/ml); KB (IC50 = 3.84 µg/ml); SK-LU-1 (IC50 = 4.56 µg/ml); SK-Mel-2 (IC50 = 3.91 µg/ml) and SW480 (IC50 = 4.65 µg/ml), respectively. The soft corals in Sarcophyton group, Sinularia group, and Menella woodin had IC50 as lower, those samples have potential sources in the field of anticancer research;
- 64 compounds were isolated from 10 selected soft corals, including 10 biscembranoid compounds (two new compounds as sarcophytolide N and M), and 19 sesquitecpen and cembranoid compounds (four new compounds as (1S,2R,8S,10R)-1,8-epoxy-2-hydroxy-guaian-3,5,7-trien-12,8-olide; (1R,2S,8S,10R)-1,2-dihydroxy-8-methoxy-guaian-3,5,7-trien-12,8-olide; Menelloide F and Menelloide G). and 30 steroid compounds (08 new compounds as menellsteroid E–H, sarcopanol A, 7-oxogorgosterol; 16α-hydroxysarcosterol and (24S)-ergostane-3β,5α,6β,25ξ,26-pentanol 26-acetate) and 5 other compounds;
- The bioactivity assays show that biscembranoid, cembranoid and steroid are potential sources in depth research on the anticancer field.
Applied results: - 10 processes for isolating 10 bioactivity compounds from the Soft corals in Northeast Vietnam sea were built. The process of isolation sarcophyton M was accepted the valid patent (Decision No. 5506/QĐ-SHTT, on 9/9/2015 accepted the valid patent)
- Trial product named SPMW7525 with 75 percent of biscembranoid enriched from soft coral Sarcophtyton pauciplicatum and 25 percent of steroid enriched from soft coral Menella woodine were built. The lethal dose (LD50) of the test product was 11.2 g per kgP. With the experiment dose around (100-200 mg per kgP), this trial product were studied on the subject of semi-chronictoxicity of the trial products in rats and rabbits, the result showed that the trial product did not affect the organs liver and kidneys of the test subjects in the study doses

| Novelty and actuality and scientific meaningfulness of the results |
- 18 new compounds were found including two new compounds as sarcophytolide N and M, four new sesquiterpene compounds as (1S,2R,8S,10R)-1,8-epoxy-2-hydroxy-guaian-3,5,7-trien-12,8-olide; (1R,2S,8S,10R)-1,2-dihydroxy-8-methoxy-guaian-3,5,7-trien-12,8-olide; Menelloide F and Menelloide G; eight new steroid compounds as menellsteroid E–H, sarcopanol A, 7-oxogorgosterol; 16α-hydroxysarcosterol and (24S)-ergostane-3β,5α,6β,25ξ,26-pentanol 26-acetate.
- The soft coral Sarcophyton pauciplicatum and Menella woodin with biscembranoid and steroid compounds have activity against several human carcinoma cells (MCF7, LNCaP, HL-60, Hep G2, KB; SK-LU-1; SK-Mel-2, SW480) were found, those soft corals are potential sourcesin depth research on the anticancer field.
