Goal and objectives of the project

- Assessment of the level of diversity and biological characteristics of symbiotic bacteria in the sponge
- Detection of strains with activity against pathogenic microorganisms

Main results

* Scientific results:
1. Survey the natural characteristics and environmental conditions of the study and sampling area of sponge (Hai Van - Son Cha).
2. Project collected 34 samples of sponges from Hai Van - Son Cha and isolated 9600 strains of attached bacteria, 9960 strains of symbiotic bacteria, 360 strains of attached actinomycetes and 940 strains of symbiotic actinomycetes, 1710 strains of attached fungi and 1462 strains of symbiotic fungi.
3. Screening 41 strains of antimicrobial bacteria and 11 strains of antimicrobial actinomycetes, including 9 bacterial strains which were able to resist more three pathogens microorganisms with a diameter of 10 mm and 1 actinomycetes strain had capable of inhibiting the growth of all three pathogens with a diameter from 14 to 17.5 mm.
4. Tested the physiological biochemical characteristics of strains selected. These all isolates were Gram (+), and their biochemical characteristics were similar to genus Bacillus.
5. Identified 16 bacterial species by using 16S rRNA gene sequencing, including 11 species of the genus Bacillus, 2 species of the genus Pseudoalteromonas and one species of the genus Sulfitobacter.
6. Identified 2 samples of sponge by using 28S rRNA gene sequence: LC25 was identified as Cliona viridis and LC28 was identified as Spongia agaricina.
7. Assessment of diversity level of sponge associated microorganisms from sponge (LC25) by using 16S Metagenomics MiSeq Sequencing. Results of the analysis showed that LC25 associated microorganisms composed of 3 kingdom, 19 phyla, 57 classes, 115 orders, 246 families, 626 geneus and 1293 species.
8. Study on the suitable culture conditions for growth and activity of two strains P. porphyrae and B.megaterium: medium with high yeast and peprone, 30°C, pH 7, salty concentration of 2-3% and a 72-hour culture period.
9. Collected 5,2g crude extracts from 50 culture liters of Bacillus megaterium and collected 3 pure compounds from the crude extracts: compound 1 (6.2 mg), compound 2 (117.8 mg) and compound 3 (49.8 mg).
10. Assessed the microbial pathogens resistant capable of crude extracts of strains LC10cs2, LC3cs2 and of 3 pure compounds from crude extracts of LC3cs2.
11. Determined the structure of the three pure compounds from crude extracts of strain LC3cs2, include 77-bis (3-indolyl) -p-cresol (1), cyclo - ((S) -Pro- (R) -Leu) (2) and Cyclo - ((S) -Pro- (R) -Val) (3).

Novelty and actuality and scientific meaningfulness of the results

Project collected 34 samples of sponges from Hai Van - Son Cha for the isolation and screening of microorganisms having capable of against microbial pathogens; study the biochemical characteristics and identify of selected microorganisms. Besides, project identified two sponges and evaluated diversity of sponge associated microorganisms by using16S Metagenomics MiSeq Sequencing.
Project also studied the suitable culture conditions for growth and express antimicrobial activity of microorganisms, thereby building a procedure of extracting bioactive substances from the culture solution of selected bacteria and assessing the antimicrobial activity of these extracts, as well as determination the structure of the extracted active substances.
