| Goal and objectives of the project | - Establish a process for obtaining ulvan-type oligosaccharide from two green algae species, Ulva lactuca and Ulva papenfussii, using an enzymatic method. |
| Main results | - Theoretical results: + Extraction, purification, and fractionation of ulvan from the green algae Ulva lactuca and Ulva papenfussii. + Determination of the chemical composition of ulvan and ulvan-derived oligosaccharides from U. lactuca and U. papenfussii including: rhamnose, iduronic acid, glucuronic acid, sulfate, xylose, glucose, and galactose. + Structural characterization of ulvan-type oligosaccharides from U. lactuca and U. papenfussii. Three structural forms were identified: A3s [→4)-β-D-GlcAp-(1→4)-α-L-Rhap3S-(1→]n; (B3s) [→4)-α-L-IdoAp-(1→4)-α-L-Rhap3S-(1→]n; and (U3s) [→4)-β-D-Xylp-(1→4)-α-L-Rhap3S-(1→]n. + Antioxidant activity of ulvan and ulvan-type oligosaccharides from U. papenfussii and U. lactucawas demonstrated through total antioxidant capacity, DPPH radical scavenging, and ferric reducing ability. + Antimicrobial activity: ulvan-derived oligosaccharides from U. lactuca and U. papenfussii showed antibacterial activity against Pseudomonas aeruginosa. Notably, oligosaccharides from U. lactuca also inhibited Escherichia coli, while ulvan from U. papenfussii did not show clear activity against this species. + Cytotoxic activity: ulvan oligosaccharide samples inhibited the growth of several tested cancer cell lines, with IC50 values ranging from 15.74 to 76.21 µg/mL. - Applied results: The findings indicate that ulvan and ulvan-derived oligosaccharides from U. lactuca and U. papenfussii represent promising materials for biomedical research and applications. |
| Novelty and actuality and scientific meaningfulness of the results | This is the first study to characterize the structure and biological activity of ulvan-derived oligosaccharides from U. lactuca and U. papenfussii produced using ulvan lyase. The study also confirmed the presence of the U3s structure in both species, which had not been previously reported. This discovery supports further work on structure–activity relationships. |
