Goal and objectives of the project

- Research on chemical composition of Alocasia odora K. Koch.
- Create a product could be inhibitor the enzyme xanthine oxidase (in vitro) and lowers uric acid in experimental mice (in vivo) to support the treatment of gout.

Main results

- Isolated and determined the structure of 07 compounds including: Alocasin A (AO1), Hyrtiosin B (AO2), β-sitosterol (AO3), β-sitosterol β-D-glucoside (AO4), Hyrtiosulawesine (AO5), myristic acid (AO6) and cis-p-hydroxycinamic acid (AO7) from the EtOAc and MeOH extracts of Alocasia odora K. Koch.
- Evaluating the inhibitory activity of XO enzyme, alpha glucosidase enzyme and NO product of extracts and compounds AO1-AO7. The test results showed that the alkaloids showed strong alpha glucosidase inhibitory activity with IC50 value from 5.25±0.32 to 29.59±3.7 µM, the inhibitory effect on XO enzyme was quite good with IC50 value from 44.34± 2.15 to 93.29±4.7 µM and moderate inhibitory effect on NO production with IC50 value 163.3±11.7 to 294.9±9.8 µM.
- Studied acute toxicity and uric acid lowering effect on experimental rats of product AO. The results showed that the dose level of 5000 mg/kg body weight did not cause death to experimental animals orally and the dose level of 500 mg/kg had the effect of lowering blood uric acid content by 26.31% compared with the disease model.
- A process has been developed to prepare AO product which is an alkaloid-rich mixture with 44.95% content, with XO enzyme inhibitory activity with an IC50 value of 18.56±1.2 µg/mL and basic standards for this product.

Novelty and actuality and scientific meaningfulness of the results

- For the first time, Alocasia odora K. Koch was evaluated for inhibitory activity of xanthine oxidase enzyme, alpha glucosidase enzyme and NO production in invitro model and uric acid lowering effect in invivo model.
